Has anyone informations and or experience with the use of an NIR-Spectroscopie system for measuring in line ?

How precise are this systems (in general, at the end of prim. drying), are there different systems ?

I am working on the development of PAT system for the lyophilization technology with FT-IR for protein biologicals. My interest towards developing the same is meant for the identification of end of primary and secondary drying points.

I am using FT-IR (Shimadzu 8400s) as a tool to develop the same.

hi Ramesh,
nice to know that you are working in development of PAT for freeze drying with FT-IR. Can you enlighten us regarding the same with some examples.

Dear Mr.Subramanium,

Thank you very much for your interest on my development. Very Happy

In general, as for as FT-IR is concerned itís going to give the results (as peaks) as per the molecule present in the composition. In case of Freeze-drying we are tend to remove the water (moisture) present in the formulation composition.

So in that case any system tool (analytical technology) is useful if it's able to detect the amount of moisture which is releasing during the process (especially during primary and secondary drying). This is the basic in this system.

Thanks for the reply. FT_IR can be used as qualitative technique. How can it be useful in measuring the mass transfer (quantitatively). If it can be done, what would be the accuracy of the values.

Dear Mr.Subramanium,

Thanks much for your elaborated inquiry.

By this system, i am not intending to quantitate the amount of moisture present in my system. By way of monitoring the moisture flow during drying precisely at the end of primary & secondary drying, we can predict the end of primary drying and the same manner for secondary drying too.

For example, if primary drying is completed that means there is no free-moisture available in the system. We can confirm the same by on-line monitoring the system with FT-IR.

Please let me know further details needed!
